SD/-Leu with Agar - Introduction
Nature:
SD/-Leu with Agar is a solid medium containing agarose, which can solidify the medium for primary cell growth. The medium contained glucose, inorganic salts, faciacic acid, other components lacking leucine and amino acids. It usually adds the corresponding antibiotic to selectively cultivate cells with the corresponding gene.
Use:
SD/-Leu with Agar is widely used in molecular genetic screening of yeast LEU2 gene mutations in yeast research. The medium can be used to screen yeasts with LEU2 gene knockout mutations and provide the required nutrients to support breeding.
Preparation Method:
The method for preparing SD/-Leu with Agar is as follows:
1. Prepare SD/-Leu liquid medium, including glucose, inorganic salts, facial acid and other components lacking leucine.
2. Add SD/-Leu liquid medium to an appropriate amount of agarose and mix well.
3. Sterilize the mixture by high temperature and high pressure sterilization.
4. Pour the sterilized mixture into a petri dish or test tube and allow it to solidify.
Safety Information:
The preparation and use of SD/-Leu with Agar requires compliance with laboratory safety practices. Agarose is flammable, keep away from open flames and high temperature sources. At the same time, antibiotics may be added to the medium, if necessary, please follow the corresponding safety instructions. Ensure proper disposal of media residues and waste during preparation and use to avoid contamination of the environment.
